Potent Antibacterial Prenylated Acetophenones from the Australian Endemic Plant Acronychia crassipetala

Abstract: Acronychia crassipetala is an endemic plant species in Australia. Its phytochemistry and therapeutic properties are underexplored. The hexane extract of the fruit A. crassipetala T. G. Hartley was found to inhibit the growth of the Gram-positive bacteria Staphylococcus aureus. Following bio-activity guided fractionation, two prenylated acetophenones, crassipetalonol A (1) and crassipetalone A (2), were isolated. Their structures were determined mainly by NMR and MS spectroscopic analyses. This is the first rec… Show more

“…Cell growth/survival assays. Cell growth/ survival of non-adherent K562 cells (3 × 10 3 cells per well) was determined using the CellTiter 96 AQueous One Solution cell proliferation assay kit as previously described 57 . Cell growth/survival of adherent cell lines (3 × 10 3 cells per well) was determined using a sulforhodamine B (SRB) assay as previously detailed 16 .…”

“…Moreover, enhanced research is needed to conserve the species and to obtain new sources for further natural product discovery. Acetophenones and chromenes have been discovered in 14 species of Melicope [ 11 , 14 , 15 , 16 , 17 , 27 , 51 , 52 , 53 , 54 , 55 , 56 , 57 , 58 ] as well as in some species of its close relatives Acronychia and Medicosma [ 33 , 46 , 59 , 60 , 61 , 62 , 63 , 64 ]. Due to the great variability of the acetophenones and chromenes in these genera, they may be of interest as biomarkers for chemotaxonomy.…”

“…Careful separation by reverse-phase chromatography provided pure samples of the two prenylation products 7a and 7b for biological assays. As O -prenylation is a modification found in antimicrobial natural flavonoids and acetophloroglucinols [ 37 ], the O -prenylated DHC 7b was also subjected to biological assays.…”
