Preconditioned and Genetically Modified Stem Cells for Myocardial Infarction Treatment

Abstract: Ischemic heart disease and myocardial infarction remain leading causes of mortality worldwide. Existing myocardial infarction treatments are incapable of fully repairing and regenerating the infarcted myocardium. Stem cell transplantation therapy has demonstrated promising results in improving heart function following myocardial infarction. However, poor cell survival and low engraftment at the harsh and hostile environment at the site of infarction limit the regeneration potential of stem cells. Preconditioni… Show more

“…Similarly, MSC-derived apoptotic bodies have been shown to control macrophage polarization for tissue repair ( Liu et al, 2020 ). Also, MSC preconditioning with physical or chemical stress enhances their survival when transplanted, by inducing the expression of pro-survival and anti-apoptotic factors ( Pasha et al, 2008 ; Raziyeva et al, 2020 ).…”

“…Experimental preclinical and clinical studies showed the positive impact of adult stem cells’ treatment on chronic wound healing [ 161 ]. This beneficial effect is due to the unique properties of stem cells to suppress immune response, repair injured tissue and support the local cells via the secretion of trophic and paracrine factors [ 162 ]. Various in vivo studies demonstrated the well-organized interaction between stem cell secretomes and mediators found in the wound area, which resulted in a lesser scar size and reduced inflammation as well as improved re-epithelialization of damaged tissue [ 163 ].…”

“…Among them are coculture methods, growth factors and cytokines, receptor agonists, hypoxia, autophagy and modifications in culture methods such as 3D culturing. An entirely different approach is the genetic modification of MSCs ( Figure 2 ) [ 148 , 149 , 150 , 151 ]. Genes, which are involved in the increase in cell survival rate, immunomodulation and regeneration, are modulated by genetically engineered constructs, such as viral vectors or plasmids [ 152 ].…”
