Prediction of Burkholderia pseudomallei DsbA substrates identifies potential virulence factors and vaccine targets

Abstract: Identification of bacterial virulence factors is critical for understanding disease pathogenesis, drug discovery and vaccine development. In this study we used two approaches to predict virulence factors of Burkholderia pseudomallei, the Gram-negative bacterium that causes melioidosis. B. pseudomallei is naturally antibiotic resistant and there are no clinically available melioidosis vaccines. To identify B. pseudomallei protein targets for drug discovery and vaccine development, we chose to search for substra… Show more

“…Additionally, given the susceptibility to non-β-lactam antibiotics (e.g. ERY, CHX, NOV), the Dsb system may be important for the function of other resistance mechanisms, such as efflux pumps (Hayashi et al 2000; Vezina et al 2020). Thus, we suggest that antibiotic susceptibility caused by defects in the DSB system is multifactorial.…”

“…Moreover, the ScsB and ScsC proteins share similarities with the DSB proteins DsbD and DsbC, respectively. Because they are essential for the folding of several virulence factors in the periplasm of bacteria, DSB proteins have been investigated as potential targets for drug discovery (Landeta et al, 2017;Bocian-Ostrzycka et al, 2017;Halili et al, 2015;Duprez et al, 2015;Vezina et al, 2020;Nebl et al, 2020;Furniss et al, 2022). Expanding our understanding of the related SCS proteins, including their mode of action and structural variation, may contribute to the identification of new druggable targets to fight bacterial infections.…”

“…pseudomallei based on a high number of discriminating peptides can increase the confidence in the diagnoses of melioidosis and glanders. Moreover, until now, diagnosis of these diseases is often cumbersome because of the diverse clinical manifestations, inadequacy of conventional diagnostic methods and lack of diagnostic facilities [13,14].…”

“…Melioidosis is a neglected tropical disease, which may spread, creating the risk that this disease will become endemic in more parts of the world. Furthermore, it is considered a potential biological threat for which no vaccine is available [2,13].…”
