“…We then tested the effect of irrelevant peptides spanning the AA sequence of a longer protein, MAGE-A3, a cancer-testis antigen expressed in several tumors including melanoma, lung and bladder cancers, and frequently investigated in cancer vaccine trials. 13 Interestingly, antigen-specific responses measured in the presence of 50, 100, 150, 200, 250 and up to 300 MAGE-A3 peptides were strongly correlated to the responses measured in the absence of MAGE-A3 peptide pools (R > 0.94 and p < 0.0001 for all peptide pools) (Fig. 1E).…”

“…Some tumors with low TMB respond completely. Response to immunotherapy has also been associated with intratumoral gene-based markers of T cell activation and antigen presentation 33,34 but such markers have not been introduced to practice. Triple negative breast cancer (TNBC), which comprises the majority of Basal-like tumors, is characterized by relatively high TMB.…”

“…911 Recently, Muthuswamy et al reported that intratumoral CD8 T cells in untreated prostate tumors were positively correlated with CXCL9 and CXCL10 levels in the tumor microenvironmen. 12 Consistent with this, we found that CXCL9 and CXCL10 are highly expressed in CD8-infiltrated prostate tumors following sipuleucel-T treatment.…”
