Objectives: Overexpression of human leukocyte antigen G (HLA-G) in several malignant tumors has been reported. The aim of our study was to investigate HLA-G expression in colorectal cancer tumors and determine HLA-G expression relation between clinicopathological characteristics and survival time.Methods: HLA-G expression was evaluated by immunohistochemistry (IHC) using anti-HLA-G antibody in 100 primary tumors of colorectal cancer with different stages. Results:Our results showed that 25% of the colorectal cancer tissues had positive HLA-G expression and 75% no stained with anti-HLA-G antibody. The HLA-G expression in advanced stages (III and IV) was more prevalent than those in earlier clinical stages (I and II) (P = 0.0001). Results showed that HLA-G expression can serve as an independent factor for overall survival (OS). In this study, patients with HLA-G expression had significantly shorter survival time than those with negative expressions (P = 0.023).Conclusions: HLA-G expression can serve as an independent factor for OS and its expression may be directly related to aggressive tumor behavior via escape from the host antitumor immune defense. Protein expression of HLA-G correlates with poor prognosis in colorectal cancer.

Aberrant induction of human leukocyte antigen-G (HLA-G) expression has been observed in various malignancies and is strongly associated with tumor immune escape, metastasis and poor prognosis. To date, great achievements have been made in understanding the underlying mechanisms of HLA-G involved in tumor progression. HLA-G could lead to tumor evasion by inhibition of immune cell cytolysis, differentiation and proliferation and inhibition of cytokine production, induction of immune cell apoptosis, generation of regulatory cells and expansion of myeloid-derived suppressive cells and by impairment of chemotaxis. Moreover, HLA-G could arm tumor cells with a higher invasive and metastatic potential with the upregulation of tumor-promoting factor expression such as matrix metalloproteinases (MMPs), indicating that ectopic HLA-G expression could render multiple effects during the progression of malignancies. In this review, we summarized the mechanisms of HLA-G involved in promoting tumor cell immune escaping, metastasis and disease progression. Special attention will be paid to its significance as an attractive therapeutic target in cancers.

The non-classical human leukocyte antigen E (HLA-E) expression is frequently overexpressed in tumor diseases, transplants and virus-infected cells and represents an immunomodulatory molecule by binding to the receptors CD94/NKG2A, -B and –C on NK and T cells. Due to its immune suppressive features HLA-E expression might represent an important mechanism of tumors to escape immune surveillance.While an aberrant expression of the non-classical HLA-G antigen in human renal cell carcinoma (RCC) has been demonstrated to be associated with a worse outcome of patients and reduced sensitivity to immune effector cell-mediated cytotoxicity, the expression and function of HLA-E has not yet been analyzed in this tumor entity.Higher levels of HLA-E transcripts were detected in all RCC cell lines and tumor lesions, which were tested in comparison to normal kidney epithelium. Immunohistochemical staining of a tissue microarray (TMA) using the HLA-E-specific monoclonal antibody TFL-033 recognizing the cytoplasmic HLA-E α-chain as monomer revealed a heterogeneous HLA-E expression in RCC lesions with the highest frequency in chromophobe RCC when compared to other RCC subtypes. HLA-E expression did not correlate with the frequency of CD3+, CD4+, CD8+ and FoxP3+ immune cell infiltrations, but showed an inverse correlation with infiltrating CD56+ cells. In contrast to HLA-G, HLA-E expression in RCCs was not statistically significant associated with a decreased disease specific survival. These data suggest that HLA-E overexpression frequently occurs in RCC and correlates with reduced immunogenicity.
