“…Gallery et al [13] reported a decreased production of MMP1 and MMP1 mRNA by endothelia in the decidual spiral arteries of the placenta of preeclamptic women. The deletion of guanine at position -1607 from the MMP1 promoter is reported to decrease transcription [14,15].…”

“…In cell culture, endothelial cells isolated from the decidual arteries of preeclamptic placentas produced smaller amounts of MMP1 than endothelia from normal decidua, and the MMP1 to tissue inhibitor of metalloproteinase-1 (TIMP-1) production ratio was low too. After incubation with phorbol myristate acetate, a potent angiogenic factor capable of inducing protease activity in endothelial cells, there was an increase in MMP1 mRNA which was higher in normal decidual endothelial cells than those from preeclamptic placentas [13].…”

“…Cytotrophoblasts and endothelial cells were isolated from biopsies of placenta and decidua at caesarean delivery as previously described 2,3 . Using these cells, we established cell bilayer co-culture in a membrane-insert system, using Millicell-PCF culture plate inserts, 12mm diameter (Millipore, Bedford, MA), in 24-well plates (Nunc, Roskilde, Denmark).…”

“…We have previously described a contribution from the host decidual endothelial cell population in regulating the de®cient vascular ingress of cytotrophoblasts in preeclampsia 2 . We now describe the development of a novel in vitro co-culture system which will permit more detailed examination of communication between human maternal decidual endothelial cells and invading cytotrophoblasts.…”

“…The initial stage of AM degradation is mediated by a matrix metalloprotease. Recently basal levels of the matrix metalloprotease MMP-1 were shown to be lower in cultured decidual endothelial cells from a pre-eclamptic sample compared to normal decidual endothelial cells [93]. This decrease was also maintained following stimulation with phorbol myristate acetate.…”
