Preliminary responses of 2, 4-D and BAP on callus initiation of an important medicinal-ornamental Hymenocallis littoralis plants

Abstract: Hymenocallis littoralis locally known as Melong kecil is an ornamental plant traditionally believed to have medicinal properties. This study was done to identify the best concentration and combinations of plant growth regulators (PGRs) for the initiation and production of friable callus. Four (4) weeks old plant was used in this study. Bulb was dissected and cleaned (sterilized) using standard surface sterilization technique and then cultured in semi-solid Murashige and Skoog (MS) media with combinations of 6-… Show more

“…1). In contrary to this similar combination with 13.50 µM 2, 4-D and 4.50 µM BAP resulted in 93.75% of callus induction percentage with yellow friable callus and was an ideal medium for callus induction in Hymenocallis littoralis (Jeevandran et al 2012). Similarly Manisha and Rajesh, (2013) also obtained maximum callus response of 91.2% on MS medium supplemented with 3.0 mg L -1 BAP and 0.5 mg L -1 2,4-D from leaf explants of Tecomella undulate (Sm.)…”

“…However, a concentration of 22.50 M 2, 4-D, and 9.50 M BAP gave a percentage of callus induction of 12.50%. Callus induction time was 15 days, and longer when compared to treatment with a CoCl2 elicitor (Sundarasekar et al, 2012).…”
