Preparation and Characterization of a Monoclonal Antibody Against Human B Lymphocyte Stimulator

Abstract: B lymphocyte stimulator (BLyS) is a member of the tumor necrosis factor (TNF) family. It is required for B cell development. Deregulation of BLyS was involved in the pathogenesis of B cell-related autoimmune diseases and multiple myeloma. To prepare monoclonal antibodies (MAbs) against BLyS, cDNA encoding soluble BLyS (sBLyS) was first amplified by reverse transcriptase-polymerase chain reaction (RT-PCR) with specific primers, and then inserted into a prokaryotic expression vector pET-30a. Right recombinant pl… Show more

“…Under the action of EDC, the introduced −COOH was coupled with the −NH 2 of the carrier protein BSA in the form of a monoamide bond (−CONH−) to synthesise the artificial antigen DON-BSA. For specific operation steps, refer to the EDC method described by Sun Zhen et al [ 31 ], with improvements. The detection antigen DON-OVA was prepared by the same method.…”

“…Recombinant human soluble BLyS (rBLyS) and anti-human BLyS MAb B7, (18) and recombinant human TRAIL (rTRAIL) were prepared in our laboratory. Human lymphoma cell lines (U937, Jurkat, Daudi, and Raji) and MM cell lines (XG-7 and SKO-007) were stored in our laboratory.…”
