“…Initial In our attempts to synthesize the various polydeoxyribonucleotides containing repeating trinucleotide sequences using complementary short-chain oligodeoxyribonucleotides as templates, we have found that the best yields were obtained with high concentrations of deoxyribonucleoside triphosphates (3-5 mM) and low temperature (15°) using either E. coli or M. luteus DNA polymerase. For the synthesis of more of the same DNAs with repeating trinucleotide sequences using previously prepared DNAas as template, the M. luteus DNA polymerase gave better yields of polymer than the E. coli DNA polymerase when the template had 100 base pairs or less (Ratliff et al, 1973). If an attempt was made to employ as template unfractionated polydeoxyribonucleotides which were themselves products of a previous enzymatic synthesis and consequently of high molecular weight, overall yields with either enzyme were less.…”
