Pretreatment With Risperidone Ameliorates Systemic LPS-Induced Oxidative Stress in the Cortex and Hippocampus

Abstract: Risperidone (RIS), an atypical antipsychotic has been found to show anti-inflammatory effect against lipopolysaccharide (LPS)-induced inflammation. In vitro study has revealed that RIS inhibits the LPS-induced reactive oxygen species (ROS) formation. We investigated the antioxidant effects of RIS on LPS-induced oxidative stress markers in Swiss albino mice. Ten weeks old male Swiss albino mice (30 ± 2 g) were pretreated with either distilled water (control) or RIS (3 mg/kg) for 7 days. On day 8, animals were c… Show more

“…However, on LPS-challenge, it was increased, as has been shown by others, with such a challenge inducing astrocyte-released cytokines to express microglial NOS and prime microglia to produce to generate more radicals upon activation in all brain areas except the brain stem. Increased MDA activity on LPS challenge as observed here and by others 7,49 was not evident in the DENtreated group, which cannot be explained in the current study. In the liver, on the other hand, MDA was not affected by LPS challenge in controls at the dose of 1 mg/kg b.w.…”

“…Total antioxidant capacity (TAC) was diminished ( Figures 12 and 13 ). Our results suggest that LPS-induced oxidative damage can be alleviated by the pretreatment with the G. oblongata marine extract and the effect is shown presumably by scavenging of the ROS by β-carotene among other bioactive secondary metabolites as an antioxidant by modulating the antioxidants status and lipid peroxidation by decreasing oxidative stress and increases the total antioxidant capacity, attenuating and protecting against LPS-induced inflammation as well as apoptosis [ 71 , 72 ]. In general, inflammatory activity is accompanied by inducible nitric oxide synthase, leading to the production of nitric oxide, which enhances the catalytic activity of COX2 via formation of the peroxinitrite anion.…”

“…Determination of superoxide dismutase activity . The SOD level was estimated based on a modified previous protocol [ 36 , 37 ]. In short, the reaction mixture carried 50 mM sodium phosphate (pH 7.8), 13 mM methionine, 75 mM nitroblue tetrazolium (NBT), 2 mM riboflavin, 100 mM EDTA and 2 mL of hippocampal tissue homogenate.…”
