Avian reovirus (ARV) infections of broiler and turkey flocks have caused significant clinical disease and economic losses in Pennsylvania (PA) since 2011. Most of the ARV-infected birds suffered from severe arthritis, tenosynovitis, pericarditis and depressed growth or runting-stunting syndrome (RSS). A high morbidity (up to 20% to 40%) was observed in ARV-affected flocks, and the flock mortality was occasionally as high as 10%. ARV infections in turkeys were diagnosed for the first time in PA in 2011. From 2011 to 2014, a total of 301 ARV isolations were made from affected PA poultry. The molecular characterization of the Sigma C gene of 114 field isolates, representing most ARV outbreaks, revealed that only 21.93% of the 114 sequenced ARV isolates were in the same genotyping cluster (cluster 1) as the ARV vaccine strains (S1133, 1733, and 2048), whereas 78.07% of the sequenced isolates were in genotyping clusters 2, 3, 4, 5, and 6 (which were distinct from the vaccine strains) and represented newly emerging ARV variants. In particular, genotyping cluster 6 was a new ARV genotype that was identified for the first time in 10 novel PA ARV variants of field isolates.

Background. Infectious diseases of the respiratory tract of farm animals are caused by a combination of infectious agents and predisposing factors. Parainfluenza virus type 3 is usually nominated as one of the causes. Seroprevalence and animal risk factors for PI3 infections were investigated in northern Jordan. Methods. The study involved 104 small ruminant flocks (18 sheep, 27 goats, and 59 mixed flocks sampled in northern Jordan. Indirect ELISA was used to test 678 blood samples used in this study. Flocks were identified as infected if at least 1 animal has been detected as positive by the ELISA test. Information regarding production and health management practices was collected in a questionnaire. Statistical analysis was conducted using the Statistical Package for Social Sciences software SPSS 23 (SPSS Inc., Chicago, IL, USA). Results. Flock-level and individual-level seroprevalences were 97%, 37%, and 76%, 11% in sheep and goats respectively. Multivariable logistic regression applied for production and health management practices showed significant risk factors for: Climatic zone (OR= 0.3) was decreasing risk factor for the PI3 seropositivity. Young separated from dams (OR= 4), neonatal deaths (OR= 3), and milking manual (OR= 37.5) were increasing risk factors for the seropositivity of PI3. Conclusions. PI3 virus has a high prevalence in sheep than goats and two risk factors could increase the prevalence of PI3 in both sheep and goats.

The σC Gene Characterization of Seven Turkey Arthritis Reovirus Field Isolates in Pennsylvania during 2011-2014

Seven turkey arthritis reovirus (TARV) field isolates made in our laboratory during 2011-2014 were characterized by sequencing of reovirus σC genes. Of the seven TARV field isolates, six were isolated from 3-to-17-week-old turkeys with tenosynovitis in six Pennsylvania farms and one was isolated from an Indiana turkey case submitted to our lab in 2014. This report describes the amplification and sequencing of the σC genes for genetic characterization studies on the seven TARV field isolates. Phylogenetic analysis of the sequence data of the seven TARV isolates together with recent published five TARVs detected in Minnesota (MN) and 25 avian reovirus (ARV) strains retrieved from GenBank revealed that all the seven PA TARV field isolates and five MN TARVs fit into genotyping cluster two when compared with a total of five different genotyping clusters (cluster 1-5) generated by the TARV and ARV reference strains. Comparison of amino acidsequences of the seven TARV isolates in cluster two with ARV vaccine strains (S1133, 1733, and 2048) in cluster one revealed that there were less than 60% similarity in nucleotide sequence and less than 56% in amino acid sequence between the two clusters. However the seven PA TARV isolates shared greater than 99% similarity with each other. Our research findings have indicated that the seven PA TARV field isolates and the five MN TARVs are grouped in the same genotype two, a separate genotype or virus species within the Orthoreovirus genus.
