Prevention of Influenza A(H7N9) and Bacterial Infections in Mice Using Intranasal Immunization With Live Influenza Vaccine and the Group B Streptococcus Recombinant Polypeptides

Abstract: We investigate the protective effect of combined vaccination based on live attenuated influenza vaccine (LAIV) and group B streptococcus (GBS) recombinant polypeptides against potential pandemic H7N9 influenza infection followed by GBS burden. Mice were intranasally immunized using 107 50% egg infectious dose (EID50) of H7N3 LAIV, the mix of the 4 GBS peptides (group B streptococcus vaccine [GBSV]), or combined LAIV + GBSV vaccine. The LAIV raised serum hemagglutination-inhibition antibodies against H7N9 in hi… Show more

“…For this purpose, we used mice, which are a widely used experimental model in the study of GBS infection [ 25 , 26 ]. In present study as well as in our early tests of the protective efficacy of recombinant GBS proteins the experiments were carried out on the model of outbred mice [ 27 – 29 ].…”

“…The LAIV of H7N3 subtype raised serum antibodies not only against the homologous virus but also against H7N9, which possessed the difference of 3% in the HA amino acid sequence. In the sera from mice double-vaccinated with H7N3 LAIV, serum HI titers against H7N9 were 20-40 times higher than against H7N3 (P < 0.05) [56]. At the same time, local IgA levels were higher against homologous H7N3 compared with H7N9 after vaccination with LAIV.…”

“…The ability of LAIV to induce antibodies not only to the homologous variant subtype but also to cross-reacting antibodies to antigenically different variants including HP variants was shown in several mouse studies [50][51][52][54][55][56].…”

“…For the prevention of respiratory infections, it is important to develop mucosal vaccines to stimulate not only systemic but also local humoral immunity. Previously, the immunogenicity, including local IgA formation, and protective efficacy of recombinant group B streptococcus (GBS) proteins in combination with LAIV, were demonstrated in mouse experiments after intranasal administration [2]. The protective effect of associated vaccination against double influenza and GBS infections correlated with serum and local antibody response against vaccine compounds and increased expression of type II interferon – interferon-gamma – in the lungs of mice [2].…”

The protective effect of associated vaccination against double influenza and GBS infections correlated with serum and local antibody response against vaccine compounds and increased expression of type II interferon – interferon-gamma – in the lungs of mice [2]. Unlike the adaptive immune response, which is very specific, the agents of innate immunity provide protection against a wide range of pathogens.…”
