Preventive effect of taurine against acute paraquat intoxication in beagles.

Abstract: Abstract-The continuous infusion of taurine markedly protected against paraquat (PQ)-induced oliguria in beagles. Pharmacokinetic studies revealed that taurine infusion increased the blood concentration of PQ and reduced the net content in the kidney and, to a lesser extent, in the lung. The excretion of PQ into the urine was unaltered. The infusion of glycine did not have such effects.

“…We hypothesized that the possible mechanism of the protective effects of taurine was via its antioxidative [29]. Based on other reports in another system [11,30,31], we found that taurine inhibits proinflammatory cytokine production in cerulein-induced pancreatitis. For these purposes, we assayed MDA as an indicator for oxidative tissue damage, and for IL-1ß and TNF-·, which are proinflammatory cytokines and known to be increased in acute cerulein-induced pancreatitis [24,25,32,33].…”

“…For example, taurine has been demonstrated to inhibit oxidative stress-induced lung damage ( 264 ), whereas more recently taurine supplementation has been shown to increase the activity of anti-oxidant enzymes in the lungs ( 265 ). In other cases, taurine has shown to exert anti-oxidant activity against bleomycin-induced lung injury ( 266 ) or against amiodarone ( 267 ) or paraquat ( 268 , 269 ) or sumatriptan mediated lipid peroxidation (LPO), and mitochondrial injury ( 270 ). Other studies have indicated that oral supplementation of taurine probably serves as a protective agent against damage mediated by nitrogen dioxide (NO 2 ) ( 271 ).…”

“…This has been particularly well documented in animal models of lung injury. Prophylactic administration of taurine protects lung from damage induced by inhalation exposure to NO 2 (2) or O 3 (3,4) and by intratracheal instillation of bleomycin (5), amidarone (6), or paraquat (7). All of these models of lung damage involve an influx of neutrophils followed by macrophages and production of inflammatory mediators.…”
