Principles of selective inactivation of viral genome

Abstract: Photosensitivity of infectious, haemagglutinating and neuraminidase activities of influenza virus has been determined experimentally. The photosensitivity of infectious activity of type B (strain Hongkong/8/73) influenza virus ws found to be higher than that of type A strains NIB-4 (N3N2-3), Leningrad/399/76 (H3N2-3) and NIB-6 (H1N1). The data obtained may be used for the determination of conditions of UV-irradiation for preparation of UV-inactivated antiviral vaccines.

“…A direct approach to prevent the transmission of airborne-mediated disease is inactivation of the corresponding airborne pathogens, and in fact the airborne antimicrobial efficacy of ultraviolet (UV) light has long been established 6 8 . Germicidal UV light can also efficiently inactivate both drug-sensitive and multi-drug-resistant bacteria 9 , as well as differing strains of viruses 10 . However, the widespread use of germicidal ultraviolet light in public settings has been very limited because conventional UVC light sources are a human health hazard, being both carcinogenic and cataractogenic 11 , 12 .…”

“…Inhibition of IAV NA was quantitatively assessed using the fluorescent substrate 4-methylumbelliferyl N -acetyl-α- D -neuraminic acid (MUN, Sigma-Aldrich) 69 70 71 . The 10-μl reaction mixture containing 0.1 mM MUN, 1 μM OC and 1 μl of the purified ultraviolet-inactivated virus in reaction buffer (50 mM sodium acetate, 6 mM CaCl 2 , pH 5.5) was prepared in a black 96-well plate on ice.…”
