Pro-inflammation NF-κB signaling triggers a positive feedback via enhancing cholesterol accumulation in liver cancer cells

Abstract: BackgroundHepatocellular carcinoma (HCC) develops in a complex microenvironment characterized by chronic inflammation. In recent years, cholesterol metabolic abnormalities have been implicated the importance in cancer cell physiology. This study was designed to investigate the relationship between inflammation and cholesterol accumulation in HCC cells.MethodsHuman HCC cells HepG2 and Huh7 were cultured and stimulated with lipopolysaccharide (LPS) for 24 h. The changes of HCC cells related to cholesterol metabo… Show more

“…Inflammation is closely associated with cancer development through multiple mechanisms 67 . In hepatocellular carcinoma cells, treatment with the pro-inflammatory factor lipopolysaccharide activates both cholesterol biosynthesis and uptake 68 . This effect is mediated by two components of the NF-κB signalling pathway, IKKα and TAB3, which are negatively regulated by miR-195.…”

“…CE was found to be modestly higher in HCC tissues compared to adjacent non-tumorous tissues, while FC was not changed, suggesting that their ratio is increased in the tumor (15). Cellular cholesterol synthesis is enhanced in liver cancer cells, and sterol regulatory element-binding transcription factor 2 and low-density lipoprotein receptor expression are induced (29). Higher endogenous cholesterol biosynthesis and uptake of low-density lipoprotein particles are essential for cell proliferation and survival (30).…”
