Probing the binding of Tb(III) and Eu(III) to the hammerhead ribozyme using luminescence spectroscopy

Abstract: This study compares the crystallographic nature of the G5 metal-binding site with solution measurements and gives a clearer picture of the coordination environment of this ion. These results provide one of the best characterized metal-binding sites from a ribozyme, so we use this information to compare the RNA site with that of typical metalloproteins.

This study also indicates that the pro-Rp oxygen is more important for metal binding in the Ce13d DNAzyme.…”

“…8,[16][17][18] Ln 3+ are important probes for nucleic acids. [19][20][21][22][23] They are quite effective in hydrolyzing RNA, 24 and early examples can be traced back to the cleavage of tRNA. 25 Ln 3+ inhibit the hammerhead ribozyme and the 8-17 DNAzyme, 22,26 but accelerate the leadzyme and a Mg 2+ -dependent ligase.…”

“…For example, both the 17E DNAzyme and the hammerhead ribozyme are inhibited by lanthanides. 15,17 On the other hand, the Leadzyme and a DNA-based ligase are accelerated by lanthanides. [18][19][20] All these examples suggest strong interactions between lanthanides and nucleic acids.…”

“…13,14 Second, a few lanthanides (especially Tb 3+ ) are luminescent and DNA can act as an antenna to increase their light absorption and thus emission intensity. This is useful for probing metal binding sites, 15 and for developing biosensors. 16 Third, lanthanides are hard Lewis acids and some have a similar size as Ca 2+ .…”
