Process Optimization of Nuciferine with Cellulase Auxiliary Ultrasonic-Assisted Extraction from Leaves of Nelumbo Nucifera Using Response Surface Methodology

Abstract: The cellulase auxiliary ultrasonic-assisted extraction process of nuciferine from Nelumbo nucifera leaves was optimized by response surface methodology. The extraction content of nuciferine was measured by HPLC, On the basis of single factor experiment, ethanol concentration, liquid-solid ratio, and extraction times were selected as independent variables, and the extraction content of nuciferine was selected as response value, Box-Behnken design was adopted and optimized extraction process in this experiment, … Show more

“…The sample was soaked in 0.3M HCl-Methanol solvent, 1:1, v/v, and sonicated in ambient temperature for 30 minutes. Combining enzyme-assisted extraction with UAE also improvesd performance [16]. Lu et al used respond surface methodology to find optimal extraction conditions, and the results showed that 10% cellulose, 87% ethanol concentration, 1:26 g/ml solid-liquid ratio, 3 times and 25 minutes were most effective [16].…”

Supercritical fluid extraction (SFE) uses the poor solubility of S-CO 2 to extract non polar compound, but the addition of methanol or ethanol can improve the solubility of S-CO 2 in polar compounds, which helps S-CO 2 extract polar compounds [13].…”

“…and two‐phase solvents (petroleum ether/ammonia solution 9). Various intensification processes, such as microwave‐assisted heating 10, ultrasonic‐assisted extraction 11, enzyme‐assisted extraction 12, β ‐cyclodextrin‐assisted extraction 13, have been employed to improve the extraction rate. Although the research of this aspect has certain progress at present, further research still should be intended to reduce extraction efforts in order to make the operation simpler and more cost‐effective.…”
