Production and Characterization of Bacterial Ghost Vaccine against Neisseria meningitidis

Abstract: Bacterial ghosts (BGS) are empty non-living envelopes produced either genetically or chemically. This study investigated a novel chemical protocol for the production of Neisseria meningitidis ghost vaccine using tween 80 followed by a pH reduction with lactic acid. For our vaccine candidate, both safety and immunogenicity aspects were evaluated. The ghost pellets showed no sign of growth upon cultivation. BGS were visualized by scanning electron microscopy, illustrating the formation of trans-membrane tunnels … Show more

“…Serum sample from animal groups were collected at the end of immunization and challenge experiment (24 h post-challenge). The procedure for determining serum bactericidal activity (SBA) is outlined in [ 16 , 56 , 57 ]. In short, 100 µL of S. flexneri 2b serotype ATCC 12022 adjusted at (1 × 10 6 CFU/mL, OD 600 :0.2) and added to 25 µL of sera from immunized and non-immunized challenged groups.…”

“…Vaccination has been recommended as the best approach to avoid infections from intracellular infectious pathogens [6]. The bacterial-ghost approach may provide a novel unconventional way to improve such a vaccine [7]. Bacterial ghosts (BGs) have been established against various microbial pathogens [8].…”
