Production and Characterization of Recombinant Human Neutrophil Chemotactic Factor

Abstract: A putative mature human neutrophil chemotactic factor (NCF) corresponding to the C-terminal 72 amino acids of its precursor was directly produced in Escherichia coli by recombinant DNA technology. Human NCF was present in both the soluble and insoluble protein fractions of the homogenate of host cells, and it was partially purified as a water-soluble polypeptide from both fractions, separately. The partially purified NCF preparation was highly purified to an endotoxin-free homogeneous polypeptide by means of C… Show more

“…Moreover, human IL-8 was used in mouse inflammatory models with a neutrophil influx peaking at 3 hours. 45 Our data indicate that short-term exposure to fever-like temperatures did not affect adhesion and spreading of GM-CSF-and IL-8-treated neutrophils to fibronectin but inhibited neutrophil migration. In accordance with our data, Frohlich and colleagues 46 demonstrated that heat did not prevent neutrophil adhesion to IL-1␤-activated endothelial cells.…”

“…Recombinant human IL-8 that was used in the crystallographic studies was purified from Escherichia coli (18). All of the x-ray data used for this structure determination were collected on crystals that were grown using protein recycled from the NMR experiments of Clore and Gronenborn and coworkers (6,7).…”

“…The proinflammatory cytokine interleukin 8/neutrophil-ac-tivating peptide-1 (IL-8/NAP-1)1 (2)(3)(4)(5)(6)(7), has been implicated as one important mediator of neutrophil infiltrates identified in the epidermal scales of psoriasis patients (8,9), the synovial fluid of rheumatoid arthritis patients (10,11), the colonic mucosa of individuals with ulcerative colitis (12,13), and in the lung of patients with idiopathic pulmonary fibrosis or adult respiratory distress syndrome (14, 15 (4). In addition to its proinflammatory role in recruiting neutrophils to local sites of acute and chronic inflammation, IL-8 may have some anti-inflammatory properties.…”
