Production and Purification of Novel Hypocholesterolemic Peptides from Lactic Fermented Spirulina platensis through High Hydrostatic Pressure-Assisted Protease Hydrolysis

Abstract: This research focuses on the proteolytic capacity of Spirulina platensis and their hypocholesterolemic activity via the 3-hydroxy-3-methyl-glutaryl-coenzyme A reductase (HMGR) inhibitory activity. To select suitable proteases for releasing peptides with high HMGR-inhibiting activity from S. platensis, eight commonly used commercial proteases were used in protease hydrolysis under high hydrostatic pressure (HHP, 100 MPa or 0.1 MPa) at 50 °C for 24 h. The Peptidase R group had the highest inhibitory capacity (67… Show more

“…Released α-amino group content, which represents the reactive amino acid, peptide, or protein containing a primary amine was determined with slight modifications through the methods used by Church et al 8 and Chen & Yang. 9 The reaction was conducted using o-phthaldialdehyde reagent added to the sample. The OPA adducted content was quantified by measuring the absorbance value at 340 nm and using Leu-Gly as a standard.…”

“…In vitro investigations indicated that HPP and SGQR suppressed the HMGCoAR activity, declined HMGCoAR and squalene synthases’ messenger RNA (mRNA) and protein expression (triple drop), and promoted the expression of low‐density lipoprotein receptor (LDLR) (1.2‐ to 1.8‐fold). The research led by Chen and Yang (2021) focused on the proteolytic potential of lactic fermented Spirulina platensis hypocholesterolemic peptides by the high hydrostatic pressure‐assisted (HHP; 100 or 0.1 MPa) protease (Peptidase R) hydrolysis for 6 h (HHP‐FH‐PR6) hydrolysis, via HMGCoAR inhibitory action. The capability of HMGCoAR inhibition was favorable with the IC 50 value of 2.9 μg/ml.…”

“…An ELISA was performed to evaluate the allergenic property of the treated b-LG using the method of Chen and Yang [17]. To estimate the amount of b-LG hydrolysates-IgE in the samples, each polystyrene microplate well was precoated by 50 µL of b-LG-specific IgE in phosphate buffer (20 mM, pH 7.5) and then washed three times using the buffer including 0.05% (w/v) Tween 20.…”

“…For example, the anti-inflammatory effect and antioxidant activity of phosvitin have been improved by HHP [16]. However, to date, it is still unclear how hydrolysis under HHP contributes to more effective proteolysis [17].…”
