Production of pectin lyase by Penicillium griseoroseum as a function of the inoculum and culture conditions

Abstract: Optimum activity of an extracellular pectin lyase produced by Penicillium griseoroseum in submerged culture was after 120 h using 0.1% (w/v) citrus pectin as substrate. Sucrose at 0.1% (w/v) stimulated enzyme production and citrus pectin gave the highest activity of enzyme per unit growth.

“…These species have been extensively studied physiologically, showing clear differences in their answers to the influence of inoculum age and concentration, as the effect of some cultural conditions (growth period, substrate concentration and different carbon sources) on the production of pectin lyase (PL) and polygalacturonase (PG). The P. griseoroseum PL activity was highest with 9-day-old spore, 5x10 4 spores/mL after 48 h of growth and the P. expansum PL activity with 5-day-old spore, 10 6 spores/mL after 72 h of growth (1)(2)(3)10,11,26,27). Several genetic studies have been carried out in our laboratory in order to isolate strains with enhanced pectinolytic enzyme activities.…”

“…Alternative carbon sources, such as sugar cane juice, have been assayed with the object of reducing the production costs of microbial PGs and PLs. The approach of pectinase production, using carbon sources of low cost and other substances to replace the natural inducer, pectin, has been used by several researchers (Brumano et al. 1993; Baracat‐Pereira et al.…”
