Profile of apoptotic proteins in oral squamous cell carcinoma: A cluster analysis of 171 cases

Abstract: Background: Oral squamous cell carcinoma (OSCC) is the eighth most prevalent cancer worldwide. In recent large-scale studies, by immunohistochemistry and cluster analysis, several markers were associated with patient survival in various tumors. The aim of this study was to analyze the expression profiles of 23 proteins that have been linked to the inhibition (Bcl-2, Bcl-x, Bcl-xL, Bcl-2-related protein A1, BAG-1, and survivin) and promotion (Bak, Bax, Bim/Bod, Bim-Long, Bad, Bid, PUMA, Apaf-1, caspase-2, caspa… Show more

“…12 The expression of Bax and Bcl-2 has previously been ad with induction and repression of apoptosis, respectively, in oral cancer cell lines. 13,14 The results of this study demonstrate that expression of Bax and Bcl-2 is key to the induction of apoptosis in HSC-3 cells. Any alteration in the expression, especially overexpression of Bcl-2, has been found in histological studies of epithelial dysplasia, in early carcinogenesis of oral cancer.…”

“…In addition, we found lower proliferation score, higher apoptosis score, higher metastasis-inhibiting score, and higher metastasis-promoting score in VEGF-high score subtype. Coutinho-Camillo et al defined a pro-apoptotic cluster and an anti-apoptotic cluster, indicating a correlation between apoptosis and tumor behavior [ 35 ]. Genes in metastasis-promoting signature were mostly related to the function of Neutrophil cell, Treg cell, and macrophage; genes in metastasis-inhibiting signature were mostly related to interferon regulatory family and the function of T cell, nature killer cell.…”

“…The cytochrome-c which is released into the cytosol binds to Apaf-1 and activates procaspase-9 in apoptosome. Caspase-9 activation causes activation of the caspase executor, in this case, caspase-3 will fragment the target protein including poly (ADP-ribose) polymerase resulting in cell death and cease of proliferation [19].…”
