Profiling the interactome of protein kinase C ζ by proteomics and bioinformatics

Abstract: BackgroundProtein kinase C ζ (PKCζ), an isoform of the atypical protein kinase C, is a pivotal regulator in cancer. However, the molecular and cellular mechanisms whereby PKCζ regulates tumorigenesis and metastasis are still not fully understood. In this study, proteomics and bioinformatics analyses were performed to establish a protein-protein interaction (PPI) network associated with PKCζ, laying a stepping stone to further understand the diverse biological roles of PKCζ.MethodsProtein complexes associated w… Show more

“…Functional HSP90 is required for the stability of Akt, which is a key gene of the PI3K-Akt signaling pathway and serves a crucial regulatory role in differentiation, cell cycle, transcription, translation, metabolism and apoptosis ( 63 ). PPP2CA serves critical roles in several cellular processes by dephosphorylating critical cellular molecules, such as Akt, P53, c-Myc and β-catenin ( 64 ). Notably, downregulation of PRKAA2 has been reported in breast tumors and other types of cancer ( 65 , 66 ), which suggests that it possesses tumor suppressor functions in vivo .…”

“…Those tools, working together, permitted the simulation and construction of proteomic models. Likewise, Hou and colleagues [11] have used Cytoscape™, a data visualization bioinformatics tool combined with MATLAB scripts, for data mining and to analyze interactome networks, i.e., the interaction between proteins. Similarly, to optimize the analysis of the numerous data generated by the Experion™ system, we wrote an algorithm using the programming platform MATLAB for data visualization, exploration, and hypothesis-driven biomarker research.…”

“…The upregulation of this pathway increases the expression of downstream survival targets (e.g., PKC-ζ). PKC-ζ has been shown to be involved in tumorigenesis, tissue invasion, and cancer progression through the modulation of cell migration machinery, such as Ras Homolog Family Member A (RhoA), Rac Family Small GTPase 1 (Rac1), and Epithelial Cell Transforming 2 (Ect2) (14)(15)(16).…”
