Programmable DNA-Fueled Electrochemical Analysis of Lung Cancer Exosomes

Abstract: Molecular diagnostics devoted to discover and monitor new biomarkers is gaining increasing attention in clinical diagnosis. In this work, a programmable DNA-fueled electrochemical analysis strategy is designed for the determination of an emerging biomarker in lung cancer, PD-L1-expressing exosomes. Specifically, PD-L1-expressing exosomes are first enriched onto magnetic beads functionalized with PD-L1 antibody and are able to interact with cholesterol-modified hairpin templates. Then, programmable DNA synthesi… Show more

“…The method achieved cascade amplification of the signal through the trans-cleavage reaction promoted by PER and Cas12a, which improved the detection sensitivity. The detection range of exosomes was from 10 3 to 10 9 particles.mL -1 , with a low detection limit of 708 particles.mL -1 103 (Fig. 5 D) .…”

“…Finally, abundant Fc-labeled DNA fragments were released and adsorbed onto the MOF-modified electrode, producing a strong electrochemical signal. Meanwhile, Sha et al used a cucurbit [ 7 ] uril (a macrocyclic molecule)-modified electrode to capture the released methylene blue tags through the host–guest interactions [ 182 ]. In this work, programmed-death-1-expressing exosomes were first captured by anti-programmed death-ligand 1 antibody-modified MBs and then labeled with cholesterol-modified hairpin templates.…”

“…A myriad of methods have been developed for EV PD-L1 detection. High-speed centrifugation can isolate EV PD-L1 from soluble PD-L1 protein by precipitation. , However, this is too time-consuming and cumbersome for clinical applications. Hence, a more convenient microfluidic affinity sorting method has been developed.…”
