Proline‑rich polypeptide‑1 decreases cancer stem cell population by targeting BAFF chromatin‑remodeling complexes in human chondrosarcoma JJ012 cells

Abstract: Chondrosarcoma is the second most common primary malignant bone tumor and is resistant to chemotherapy and radiation. Inadequate treatment response and poor prognosis requires novel therapeutic approaches. Proline-rich polypeptide-1 (PRP-1), synthesized by brain neurosecretory cells, has demonstrated antitumor properties in JJ012-cells; however, its underlying molecular mechanism remains unclear. The present study aimed to investigate the epigenetic regulation by which PRP-1 inhibits chondrosarcoma cancer stem… Show more

“…Tumor suppressors, such as miR20a, miR125b, and miR192, were significantly upregulated by mTORC-1 inhibitor PRP-1 while onco-miRNAs, miR509-3p, miR589, miR490-3p, and miR550 were downregulated in the human chondrosarcoma JJ012 cell line [50]. The fact that PRP-1 manifests itself as a powerful epigenetic regulator was confirmed with the experiments demonstrating inhibition of BAFF Chromatin remodeling complexes [51].…”

“…PRP1 is known to have cytotastic, antiproliferative, immunomodulatory, and tumor suppressor properties, and is an mTOR inhibitor ( Figure 1 ). It was shown that PRP1 treatment on the JJ012 cells’ monolayer and the 3D spheroid model significantly decreased the ALDH high CSC population [ 134 , 135 ]. A combination of disulfiram and Cu 2+ (DSF/Cu) has also been proven to target CSCs.…”
