Prospective Evaluation of the Chemiluminescence Test for the Detection of Granulocyte Antibodies: Comparison with the Granulocyte Immunofluorescence Test

Abstract: A series of 213 neutropenic patients were tested for the presence of granulocyte antibodies using the granulocyte chemiluminescence test (GCLT) and the granulocyte immunofluorescence test (GIFT). Sera containing lymphocyte (HLA) antibodies were excluded from the study. A direct GIFT was performed on granulocytes from 56 patients. Samples were obtained from patients with a range of clinical conditions including primary adult autoimmune neutropenia, autoimmune neutropenia of infancy, autoimmune neutropenia secon… Show more

“…6 If specific neutrophil antigens are targeted, demonstration of immunoglobulin on neutrophils or their precursors can be used diagnostically, with a sensitivity ranging from 78% to 98%. 3,4,7 Targeting of specific antigen results in opsonization of neutrophils and removal by macrophages. The actual site of removal is presumably extravascular, within spleen or marrow or both.…”

“…As an alternative/addition to GAT, the GIFT/LIFT method may be complemented by the functional granulocyte chemiluminescence test (Hadley & Holburn, 1984; Lucas, 1994) where unfixed, heat‐treated granulocytes/lymphocytes (from the same donor) are incubated with patient sera. The incubated, unfixed cells are then added to a pool of random donor monocytes which phagocytose any granulocytes with bound antibody.…”

“…PMNs were separated from citrate-anticoagulated peripheral blood samples collected from several healthy individuals by the conventional method involving Ficoll density gradient centrifugation, harvesting of the polymorphonuclear cell layer and red cell lysis in NH 4 Cl solution (Lucas, 1994). Subsequently, the cells were purified by negative selection in a mixture of magnet-conjugated anti-CD3, anti-CD4, anti-CD8, anti-CD14, anti-CD19 and anti-CD56 Abs (Dynabeads, Dynal AS, Oslo, Norway), followed by harvesting of magnet-nonadherent cells as described previously (Ochensberger et al, 1999).…”
