Proteasome Inhibition After Burn Injury

Abstract: Objective To assess the effects of proteasome inhibition on the development of burn-induced hypermetabolism. Methods Rats underwent 30–40% TBSA scald burn or sham injury. The proteasome inhibitor bortezomib (0.1 mg/kg) or vehicle (n=10) was administered i.p. 3×weekly starting at 2h (Early-Bortezomib, n=20) or 48h (Late-Bortezomib, n=13) post-burn. Body weights were determined weekly. Resting energy expenditures (REE) were measured at days 0 (baseline), 7, 14, 21 and 42 post-burn. At day 42, blood and pectora… Show more

“…All aspects of a study must be fully described in order to make it reproducible. We did find two circumstances in studies that followed Walker and Mason´s model 6 ; however, the authors did not inform if they made changes to the model 13 , 14 , 19 , 20 , 23 . In reading the articles in full, we noticed changes in temperature, location and injury extent.…”

“…The sum of the water temperature and the length of exposure time to the water are important to determine the degree of the injury. Vana et al 20 . only mention ‘boiling water’, which is relative, for the boiling point of water depends on the altitude of the environment.…”

“…Male Sprague Dawley rats (325–350g body weight, Harlan, Indianapolis, IN) were anesthetized with 2.5% isoflurane (Patterson Veterinary, Devens, MA), shaved, and placed into a template that exposes a dorsal body area corresponding to 30% of their total body surface area. A full thickness burn was then induced by immersion of the dorsal skin into boiling water for 17 seconds, as described previously (13,14). Sham animals were treated as described before, except that their dorsal surface was immersed in tepid water.…”

“…Inhibition of UPS may be a therapeutic option to prevent ICUAW. Proper timing of proteasome inhibition, however, will be crucial, since its use in pre-clinical models is associated with delay and attenuation of resting energy expenditure as well as increased mortality, specifically when implemented early after injury [ 85 ].…”
