Gold nanoparticles (AuNPs) are widely studied materials that have intense applications in various fields. Different methods use to synthesize the AuNPs, one of them is eco-friendly biological method, known as "green synthesis" a simple and inexpensive method. The Synthesis of AuNPs, using ginger extract is described here. The components found in ginger work as a stabilizing and reducing agent. Hydrogen tetrachloroaurate (HAuCl4) are mixed with the ginger extract to produce AuNPs. The Surface Plasmon Resonance (SPR) measured at λmax = 540 nm. UV-Vis, FTIR and Zeta potential are used to identify AuNPs, as well as AFM and SEM. Data indicate that AuNPs has a spherical structure. The results demonstrate that AuNPs stabilized by ascorbic acid were used as a colorimetric probe for Hg2+ ions, based on the production of (Au-Hg) amalgamate which made changes in their absorbance, due to its ability to reduce Hg2+ to Hg0, to enhance amalgamation. Practically, this procedure has successfully worked to detect Hg2+ in tap water as a sensitive and selective probe with upper limit of detection equal to 120 µM as well as the limit of detection (LOD) equal to 0.65 µM, so the method proved to be sensitive and selective probe.

Diabetes Mellitus is a metabolic disorder characterized by elevated blood glucose level caused due to deficiency of insulin secretion or insulin action. The management of the blood glucose level is an important approach in the control of diabetes mellitus complications. There are many divergent therapeutic strategies in the management of Type II diabetes. The inhibition of carbohydrate hydrolyzing enzymes such as α-amylase and α-glucosidase can be used to lower postprandial blood glucose levels. The objective of the present study is to evaluate the inhibitory potential of Gymnemic acid in various concentrations on α-amylase and α-glucosidase activities. Gymnemic acid and voglibose show the IC50 value of 68.12 and 46.23 on alpha amylase and IC50 value of 65.77 and 36.67 on alpha glucosidase activity respectively. Insilico docking study between α-amylase and α-glucosidase with gymnemic acid and voglibose depict the similar amino acid binding sites that substantiate that gymnemic acid might be used as a natural and cost-effective alternative to the synthetic drug. This study throws light on the anti-diabetic action of phytomedicine, Gymnemic acid, a component of Gymnema sylvestre.

Objectives: To evaluate the therapeutic potential of Gymnema sylvestre (GS) leaf ethanolic extract against letrozole induced polycystic ovarian syndrome (PCOS) in rats. Methods: Thirty female virgin rats (180 - 250g) with regular estrus cycles were used and divided into five groups of six animals each. Group-I serves as normal control receives 1ml of 0.5% carboxyl methyl cellulose orally; group-II to V were treated with letrozole (1mg/kg/b.w. p.o.) for 21 days to induce PCOS. Induction of PCOS was confirmed by measuring menstrual irregularity. After induction of PCOS, group-II serves as PCOS control, group-III receives metformin (20 mg/kg/b.w. p.o.) as standard, and group- IV to V treated with G. sylvestre ethanolic extract 200mg/kg/b.w. and 400mg/kg/b.w. p.o. respectively for 28 days. After completion of the treatment schedule oral glucose tolerance test was carried out, followed by serum lipid profile (LDL, VLDL, HDL, and TG), and hormonal analysis was carried out. Then the animals have sacrificed and the ovaries were then processed for histological examination. Results: The results of this study exhibited irregularity in the estrus cycle, a significant increase in serum blood glucose, and an altered serum lipid profile in PCOS control. In addition, the PCOS control rats showed a significant increase in serum luteinizing hormone, testosterone, and a decrease in estrogen, follicle-stimulating hormone, and progesterone. These changes were significantly reversed by G. sylvestre in a dose dependent manner. Histopathology of the ovary also showed an almost normal ovary in the treatment groups. Conclusion: In conclusion, the treatment of ethanolic leaf extract of Gymnema sylvestre reduces insulin resistance by improving glucose utilization, regularize menstrual irregularity and promote ovulation by correcting the hormonal irregularities and improve the development of follicles in PCOS condition.
