Protective Effects of 2′,4′-Dihydroxy-6′-methoxy-3′,5′-dimethylchalcone to PC12 Cells against Cytotoxicity Induced by Hydrogen Peroxide

Abstract: Oxidative stress has been considered as a major cause of cellular injuries in various clinical abnormalities. One of the possible ways to prevent reactive oxygen species (ROS)-mediated cellular injury is dietary or pharmaceutical therapies to augment the endogenous antioxidant defense capacity. The present study found that 2',4'-dihydroxy-6'-methoxy-3',5'-dimethylchalcone (DMC), a chalcone isolated from the buds of Cleistocalyx operculatus, possessed cytoprotective activity in PC12 cells treated with H(2)O(2).… Show more

“…The compound DMC has been isolated from C. operculatus [7], M. serrata [8], and S. samarangense [9]. DMC possessed hepatoprotective [10], cytoprotective [11], anti-inflammatory [12], antifungal, antibacterial [8], anti-TB [13], antiviral [14], antispasmodic [15], antidiarrhoeal [16], antihyperglycaemic [17], anti-MDR [18, 19], and antiapoptotic effect [20]. DMC has been evaluated for its cytotoxic activity on various human cancer cells including liver cancer (SMMC-7721s), HeLa (SPC-A-1, 95-D, and GBC-SD) [7], colon (SW-480) [9], leukemia (K562) [21], colorectal carcinoma (HCT-116 and LOVO) [22], breast (MDA-MB-453), and ovarian [23] cell lines.…”

“…ROS causes selective degeneration of oligodendrocytes (Griot et al, 1990) by promoting LPO (Su et al, 2011). Following a hypoxic-ischemic injury, the white matter showed increased levels of MDA ) and 4-HNE, which are toxic to axons and oligodendrocytes (Lin et al, 2004).…”

“…GSH depletion in oligodendrocytes is implicated in their susceptibility to free radical-mediated injury (Back et al, 1998). Activated microglia have been reported to release ROS (Block et al, 2007), which induces LPO (Su et al, 2011). On the basis of these observations, the primary cultured oligodendrocytes were divided into six groups and treated as described below for GSH and LPO measurement.…”

“…Photodynamic FA-TiO 2 could result in subsequent cell damage and a salient feature is the decrease in MMP [33,34], which was monitored by Rh 123 fluorescence. As shown in Fig.…”
