Frequent and improper use of herbicides disrupts a plant's metabolism, causing oxidative stress that degrades crop quality. However, few studies have examined the inhibitory effects of herbicides on plant growth and defense mechanisms in terms of their impact on soil quality and crop rhizosphere. Therefore, the current study investigated the detrimental impacts of six typical and multilevel herbicides on the microbial community and signal molecules in the soil as well as on the levels of hormones and secondary metabolites in wheat seedlings. Interestingly, bensulfuron-methyl, terbutylazine (TBA), and 2,4-D butylate significantly induced oxidative damage while reducing the number of phytohormones (salicylic acid and jasmonic acid) and secondary metabolites (tricin, quercetin, and caffeic acid) in the roots and leaves compared with the controls, isoproturon, fenoxaprop-p-ethyl, and pretilachlor. At twice the recommended levels (2×), they also decreased the microbial α diversity and, in particular, the abundance of Gammaproteobacteria, Alphaproteobacteria, Actinobacteria, Bacteroidia, Verrucomicrobia, Bacilli, Acidimicrobiia, Deltaproteobacteria, and Gemmatimonadetes by disrupting the level of enzymes (e.g., urease and sucrase) and metabolites (indole-3-acetic acid, salicylic acid, apigenin, 4-hydroxybenzoic acid, DIMBOA, and melatonin) in the rhizosphere soil. Overall, significant exposure to herbicides may inhibit wheat growth by disturbing the microbial composition in the rhizosphere soil and the distribution of secondary metabolites in wheat seedlings.

Flavonoids, structured by the skeleton of phenyl benzopyrone, are widely occurring polyphenolic compounds in plants (Maleki et al., 2019). As common substances in the human diet, flavonoids are associated with numerous health benefits. Studies prove that a moderate habitual intake of flavonoids can reduce all-cause, cancerand cardiovascular-related mortality (Bondonno et al., 2019). The flavonoid quercetin and rutin (quercetin-3-Orhamnosylglucoside) (Figure 1) exhibit multiple bioactivities, including antioxidative, anti-inflammatory, antiviral, antidiabetic, antitumor, antibacterial, and neuroprotective activities (Gullón et al., 2017). Due to their excellent beneficial effects on human health, rutin and quercetin are popular dietary supplements (Zhang et al., 2017). The application areas of quercetin and rutin are not only restricted to the food industry, they are also used in cosmetics, for example, as an ingredient in sunscreen, and pharmaceutical industries (Berlier et al., 2013). Credible and simultaneous determination of multiple pesticides is highly desirable for guaranteeing food safety. However, the current methods are limited to significant interference of color and fluorescence or electrode's modification and mainly focus on the analysis of a single pesticide. Herein, we proposed a novel aptamer-based homogeneous electrochemical system for highly sensitive and simultaneous analysis of multiple pesticides based on target pesticide-switched exonuclease III (Exo III)-assisted signal amplification. The recognition of hairpin probes by target pesticides impels the production of pesticide−DNA complexes, which hybridize with electroactive dye-labeled DNA to form double-stranded DNA, subsequently initiating an Exo IIIassisted digestion reaction to generate abundant electroactive dye-tagged mononucleotides. In comparison with pesticide deficiency, two higher differential pulse voltammetry (DPV) currents are measured, which rely on the amount of target pesticides. Therefore, simultaneous analysis of two pesticides is realized with limits of detection of 0.0048 and 0.0089 nM, respectively, comparable or superior to those of known methods that focused on a single pesticide. Moreover, the proposed system is successfully employed to simultaneously evaluate the residual level of acetamiprid and profenofos in Brassica chinensis and thus will find more useful applications for pesticide-related food safety.
