Protective mechanism of artemisinin on rat bone marrow-derived mesenchymal stem cells against apoptosis induced by hydrogen peroxide via activation of c-Raf-Erk1/2-p90rsk-CREB pathway

Abstract: BackgroundBone marrow-derived mesenchymal stem cell (BMSC) transplantation is one of the new therapeutic strategies for treating ischemic brain and heart tissues. However, the poor survival rate of transplanted BMSCs in ischemic tissue, due to high levels of reactive oxygen species (ROS), limits the therapeutic efficacy of this approach. Considering that BMSC survival may greatly enhance the effectiveness of transplantation therapy, development of effective therapeutics capable of mitigating oxidative stress-i… Show more

“…Conversely, a shift in this balance toward Bax evokes apoptosis. [ 91 ] Bcl‐2 performs a vital protective role against oxidative stress‐induced cell death via diverse mechanisms. [ 92,93 ] Therefore, the robust expression of Bcl‐2 and mild expression of Bax suggest that the administration of Cu to rats supports proliferation in the brain tissue.…”

“…A mass of evidence suggests that PI3K/AKT and ERK1/2 signaling pathways are related to cell proliferation and apoptosis [ 10 12 ]. Ascending expression of AKT phosphorylation and ERK1/2 phosphorylation seems to be pivotal anti-apoptotic signals [ 13 , 14 ]. Moreover, Caspase 3 acts as the key executor of apoptosis [ 15 ].…”

“…Mitochondrial bioenergetics is closely associated with a normal MMP level, which is important for ROS scavenging and cytoprotection against apoptosis induced by excessive ROS [ 30 , 31 ]. The fluorescent lipophilic cation JC-1 is an MMP-sensitive dye that aggregates in the mitochondrial matrix.…”

“…The fluorescent lipophilic cation JC-1 is an MMP-sensitive dye that aggregates in the mitochondrial matrix. It exhibits red fluorescence in healthy cells and emits green fluorescence as a monomer [31]. As shown in Fig.…”
