Proteome-Level Analysis of Metabolism- and Stress-Related Proteins during Seed Dormancy and Germination in Gnetum parvifolium

Abstract: Gnetum parvifolium is a rich source of materials for traditional medicines, food, and oil, but little is known about the mechanism underlying its seed dormancy and germination. In this study, we analyzed the proteome-level changes in its seeds during germination using isobaric tags for relative and absolute quantitation. In total, 1,040 differentially expressed proteins were identified, and cluster analysis revealed the distinct time points during which signal transduction and oxidation-reduction activity chan… Show more

“…In the present study, three heat shock proteins, including VIT_08s0007g00130.t01 (heat shock cognate 70 kDa protein 2), VIT_16s0050g01150.t01 (heat shock protein 83), and VIT_01s0010g00680.t01 (heat shock protein 90-5, chloroplastic), exhibited differential expression pattern in the HC-treated buds as compared to those of control (Table 1). A previously published study illustrated that the heat shock proteins protected against oxidative injury in beech seeds germination (Chang et al 2018). Therefore, the results of this study depicted that metabolism-related proteins might be involved in grape bud dormancy release after HC application.…”

“…Proteins associated with protein synthesis are required for cell division and can be utilized as a signaling molecule for meristematic tissue state (Chang et al 2018). In the current study, the elongation factor 1-alpha showed higher expression in the HC-treated bud samples, which indicated the involvement of this protein in the dormancy release of grapevine after the HC application.…”

“…Several studies have investigated proteomic, transcriptomic, or metabolic changes during either cold stratification or after-ripening in Prunus campanulata ( Lee et al, 2006 ), A. thaliana ( Angelovici et al, 2011 ; Arc et al, 2012 ), Acer pseudoplatanus ( Pawłowski and Staszak, 2016 ), Gnetum parvifolium ( Chang et al, 2018 ), and Arachis hypogaea ( Xu et al, 2020 ). The mechanism of seed dormancy break is thus well studied and understood.…”

“…Several studies have been conducted to explore the changes in the proteome or metabolome during either cold stratification or after-ripen for the determination of metabolic activities involved in seed germination mechanisms (Lee et al, 2006; Angelovici et al, 2011; Arc et al, 2012; Pawłowski and Staszak, 2016; Chang et al, 2018). It is the metabolic activities occurred before radical protrusion that are responsible for the delay or no germination of seeds (Bewley et al, 2013).…”
