Heterostyly is a common floral polymorphism, but the proteomic basis of this trait is still largely unexplored. In this study, self- and cross-pollination of L-morph and S-morph flowers and comparison of embryo sac development in eggplant (Solanum melongena L.) suggested that lower fruit set from S-morph flowers results from stigma-pollen incompatibility. To explore the molecular mechanism underlying heterostyly development, we conducted isobaric tags for relative and absolute quantification (iTRAQ) proteomic analysis of eggplant pistils for L- and S-morph flowers. A total of 5,259 distinct proteins were identified during heterostyly development. Compared S-morph flowers with L-morph, we discovered 57 and 184 differentially expressed proteins (DEPs) during flower development and maturity, respectively. Quantitative real time polymerase chain reactions were used for nine genes to verify DEPs from the iTRAQ approach. During flower development, DEPs were mainly involved in morphogenesis, biosynthetic processes, and metabolic pathways. At flower maturity, DEPs primarily participated in biosynthetic processes, metabolic pathways, and the formation of ribosomes and proteasomes. Additionally, some proteins associated with senescence and programmed cell death were found to be upregulated in S-morph pistils, which may lead to the lower fruit set in S-morph flowers. Although the exact roles of these related proteins are not yet known, this was the first attempt to use an iTRAQ approach to analyze proteomes of heterostylous eggplant flowers, and these results will provide insights into biochemical events taking place during the development of heterostyly.

Background: Leaf senescence is one of the most common manifestations in plant senescence and has an important effect on plant photosynthesis. Delaying or shorten the senescence of tobacco leaves is one of the important approaches to improve tobacco breeding. However, the molecular regulatory mechanism of tobacco leaf senescence in response to photosynthesis is still poorly understood. To gain insights into the senescence of tobacco leaves, we integrated analyses including photosynthesis, organelle ultrastructure, and proteome of tobacco leaves during senescence. Results:The photosynthetic rate, intercellular CO 2 concentration, stomatal conductance and transpiration rate showed a downward trend, and the stability of organelle decreased with the senescence of tobacco leaves. Isobaric Tag for Relative Absolute Quantitation (iTRAQ) and Parallel Reaction Monitoring (PRM) were used to analyze the proteins expressed in different periods that were estimate based on photosynthetic physiology and ultramicroscopic observations. A total of 321, 319, 223 differentially expressed proteins (DEPs) were identified from over maturity (OM) vs immature (IM), OM vs well maturity (WM) and WM vs IM, respectively, including 122/199, 124/195 and 125/98 up/down proteins, respectively. Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis revealed that the DEPs were significantly enriched in metabolic pathways, biosynthesis of secondary metabolites, microbial metabolism in diverse environments and starch and sucrose metabolism. In addition, the down-regulated proteins were also involved in metabolic pathways, such as carbon sequestration of photosynthetic organisms and photosynthesis. PRM analysis was performed and indicated that iTRAQ is highly reliable in the identification of major proteins involved in tobacco senescence. Conclusions:This study provided important technical references for screening of host photosynthetic proteins during tobacco leaves senescence and established a basis for scientifically understanding of the senescence mechanism of tobacco leaves.

The aim of this study was to investigate the patterns of antibiotic resistance of Aeromonas spp. bacteria isolated from the sea trout (Salmo trutta) from the state fish hatcheries of the Institute of Food Safety, Animal Health and Environment 'BIOR', Latvia. Bacteriological investigations were performed at four state fish hatcheries located in the drainage basins of the main Latvian rivers -Daugava, Venta and Gauja, during the five-year period (2012 -2016). In fish with visible clinical signs, bacteriological samples were collected from heart, liver, spleen, kidney and ulcer surfaces. Aeromonas hydrophila and Aeromonas salmonicida were isolated from sea trouts. A total of 52 individual sea trouts were examined. Resistance to amoxicillin, ampicillin, cephalexin, colistin, doxycycline, enrofloxacin, erythromycin, florfenicol, gentamycin, kanamycin, lincomycin, neomycin, oxytetracycline, spectinomycin, streptomycin, tetracycline, trimethoprim/sulfamethoxazole was tested. The results of this study suggest a multi-drug resistance pattern among the A. hydrophila isolates. All the isolates were resistant to amoxicillin (100%), ampicillin (100%), cephalexin (100%) and erythromycin (100%). The lowest level of resistance was found against florfenicol (4.55%), gentamycin (4.55%), kanamycin (4.55%), but susceptibility was recorded to enrofloxacin, neomycin and trimethoprim/sulfamethoxazole. A. salmonicida isolates were resistant to oxytetracycline (9.38%) and tetracycline (9.38%). For other antibiotics A. salmonicida isolates were susceptible.
