Proteomic Profiling for Metabolic Pathways Involved in Interactive Effects of Elevated Carbon Dioxide and Nitrogen on Leaf Growth in a Perennial Grass Species

Abstract: Elevated atmospheric CO2 and nitrogen are major environmental factors affecting shoot growth. The objectives of this study are to determine the interactive effects of elevated CO2 and nitrogen on leaf growth in tall fescue (Festuca arundinacea) and to identify major proteins and associated metabolic pathways underlying CO2-regulation of leaf growth under insufficient and sufficient nitrate conditions using proteomic analysis. Plants of tall fescue treated with low nitrate level (0.25 mM, LN), moderate nitrate … Show more

“…Total protein of 8 h imbibed seeds was extracted using phenol-based method and the concentrations were measured with bovine serum albumin. Next, 100 μg protein was used for iTRAQ labelling proteomics analysis [ 55 ]. Protein digestion and labelling were performed according to the manufacturer's instructions (AB SCIEX, Concord, ON).…”

“…Data acquisition was performed with a TripleTOF 5600 mass spectrometer (MS) (AB SCIEX, Concord, ON) fitted with a Nanospray III source (AB SCIEX, Concord, ON). Data were acquired on the parameters as previously reported [ 55 ]. The original mass data were processed with ProteinPilot 5.0 (AB SCIEX, Concord, ON) and searched against the Uniprot protein database ( http://www.uniprot.org ) and G. hirsutum TM-1 protein FASTA ( https://www.cottongen.org/ ) [ 56 ].…”

“…A large number of studies reported that elevated CO 2 could promote plant growth and development, including perennial grass species, such as tall fescue ( Festuca arundinacea Schreb.) [ 13 , 14 , 15 , 16 ], Kentucky bluegrass ( Poa pratensis L.) [ 17 , 18 ], and bermudagrass ( Cynodon dactylo n (L.) Pers.) [ 19 , 20 ].…”
