Protracted Protection to Plasmodium berghei Malaria Is Linked to Functionally and Phenotypically Heterogeneous Liver Memory CD8+ T Cells

Abstract: We previously demonstrated that protection induced by radiation-attenuated (γ) Plasmodium berghei sporozoites is linked to MHC class I-restricted CD8+ T cells specific for exoerythrocytic-stage Ags, and that activated intrahepatic memory CD8+ T cells are associated with protracted protection. In this study, we further investigated intrahepatic memory CD8+ T cells to elucidate mechanisms required for their maintenance. Using phenotypic markers indicative of activation (CD44, CD45RB), migration (CD62L), and IFN-… Show more

“…Th1-type responses also control Leishmania major but Th2-type responses exacerbate the disease [27]. This is consistent with our observation [7] that IFN-c-producing liver CD8 T cells increase between 1 and 6 h after challenge of cspz-immune mice. Effector CD8 T cells also might require costimulation by CD40 and/or CD80, which increased on c-spz-immune KC after challenge; moreover, they might rely on activated CD4 T cells, thus it is relevant that class II molecules on KC increased, albeit insignificantly, after challenge of c-spz-immune mice.…”

“…MHC class I and inflammatory cytokines are sine qua non for the activation of Ag-specific effector CD8 T cells, which are not detected in spz-challenged naive mice [7]. Spz may have interacted directly or indirectly with KC.…”

“…Naive T cell expansion and conversion into conventional memory T cells, as well as central (proliferation) and effector (IFN-c secretion) memory T cell effector functions were also affected. In liver-stage malaria, protective immunity can be associated with specific central or effector memory T cell subpopulations [17,18]. The existence and selection of these novel multi-level (naive, memory-central and memory-effector) antagonistic T cell epitopes within MSP-1 therefore offers an unusual and powerful immune evasion strategy for the malaria parasite.…”
