Prussian Blue Nanozymes with Enhanced Catalytic Activity: Size Tuning and Application in ELISA-like Immunoassay

Abstract: Prussian blue nanozymes possessing peroxidase-like activity gather significant attention as alternatives to natural enzymes in therapy, biosensing, and environmental remediation. Recently, Prussian blue nanoparticles with enhanced catalytic activity prepared by reduction of FeCl3/K3[Fe(CN)6] mixture have been reported. These nanoparticles were denoted as ‘artificial peroxidase’ nanozymes. Our study provides insights into the process of their synthesis. We studied how the size of nanozymes and synthesis yield c… Show more

“…have been reported for enzymes, only a few papers discuss these modifications for nanozymes. [24][25][26] Finally, the selection of the substrate composition should also consider the stability of the components. While for some nanozymes, high concentrations of substrates may be favourable, the applicability of such substrate solutions will be limited by the high background caused by the self-oxidation of substrates.…”

“…Therefore, it is necessary and urgent to develop suitable contrast agents with high sensitivity and affinity for malignant tissues for molecular imaging. Currently, many researchers are devoted to improving the detection sensitivity of nanozymes and optimizing the design of nanozyme-like catalytic activity, including size adjustment, composition, heteroatom doping, and specific surface modifications [ 80 , 81 , 82 , 83 , 84 ]. Nanozymes recognize some specific TME biomarkers to improve the diagnostic efficacy of early cancer, such as abnormal fluctuations and expression of metal ions and miRNA [ 85 , 86 ].…”

“…Prussian Blue nanoparticles were synthesized by reducing FeCl 3 /K 3 [Fe(CN) 6 ] with H 2 O 2 in the presence of citric acid [29]. Then, the nanoparticles were coated with a gelatin A layer, to which streptavidin (PB@Gel/Str) or BSA (PB@Gel/BSA, control nanoparticles) was covalently attached.…”
