Based on the codon preference of Escherichia coli, the gene of lipase from Schizochytrium pombe was optimized and the codon adaptation index was 0.8 and the gene was expressed in Escherichia coli as a His-tag fusion protein using pET-30a as the expression vector. The recombinant strain with OD600 about 0.6 was induced with 1 mmol/L isopropyl-β-D-thiogalactoside at 30 °C for 12 h. The purified recombinant lipase STGL3 had a molecular mass of 32 kDa, and the purified lipase had a specific enzyme activity of 2.29 μmol/(min•mg), which was 2.3 times higher than before, and a protein content of 650.00 mg/L. The recombinant STGL3 showed the highest hydrolysis activity at 40 °C and pH 7.5, respectively. The effect of different metal ion concentrations on enzyme activity varied, lipase STGL3 was stable in the presence of Ca2+, while Mg2+, Mn2+, Zn2+, Fe2+ and EDTA had an inhibitory effects on lipase activity. Different types of surfactants and organic solvents inhibited the enzymatic activity of recombinant lipase to different extents. The recombinant lipases showed a preference towards pNP-esters with medium and longer acyl-chains. (C12, C14 and C16) and the recombinant lipases showed relatively weak lipase activity towards the shorter carbon chains. The kinetic constants of lipase were 0.29 mmol/L for Km, 2.28 mmol/(L•min-1 ) for Vmax, and 6.19 S-1 for Kcat. The present study not only achieved the expression and characterization of enzymatic properties of lipase from Leptosphaeria japonica, but also provided some basis for the study of enhancing the quality and quality of algal oil by means of genetic engineering, and further exploring the potential industrial applications of Leptosphaeria japonica lipase.

Summary Rice grains were obtained from two Chinese provinces (Hubei and Guangxi) and then their rice bran was abraded into four equal fractions by weight from outside to inside. The component distributions in these four fractions were then investigated. The contents of total lipid‐soluble antioxidant showed no significant difference between rice bran from the Hubei province and the Guangxi province. But the content of total water‐soluble antioxidant was 1690.20 ± 86.40 mg kg−1 for rice bran from the Hubei province, significantly higher than those from the Guangxi province (1014.67 ± 71.09 mg kg−1). Obvious differences between four rice bran fractions or two provinces were proved by principal component analysis (PCA). Lipase activities decreased significantly from outer fractions to inner fractions, and lipids mainly distributed in three outer fractions. The essential element contents decreased significantly, but a main heavy metal element (Pb) in cereal increased significantly to 0.36 mg kg−1 from outer fractions to inner fractions, which was just slightly below the upper limit of Chinese Standard (0.40 mg kg−1). Although protein contents had no significant difference in four fractions, the amino acid compositions were quite different. In summary, different fractions of rice bran had different compositions, which could provide guidance for the classified processing of rice bran.

Rice bran is a valuable byproduct from the food processing industry, which contains abundant protein, essential unsaturated fatty acids, and numerous bioactive compounds. However, its susceptibility to rancidity greatly restricts its wide utilization. Many strategies have been proposed to delay the rancidity of rice bran, but most of them have their respective limitations. Here, we proposed that developing rice ban lipase peptide inhibitors represents an alternative and promising prescription for impeding the rancidity of rice bran, in contrast to the conventional stabilization approaches for rice bran. For this reason, the rancidity mechanisms of rice bran and the research progress of rice bran lipases were discussed. In addition, the feasibility of utilizing in silico screening and phage display, two state-of-the-art technologies, in the design of the related peptide inhibitors was also highlighted. This knowledge is expected to provide a theoretical basis for opening a new avenue for stabilizing rice bran.
