Qualitative and Quantitative Neuropathology Approaches Using Magnetic Resonance Microscopy (Diffusion Tensor Imaging) and Stereology in a Hexachlorophene Model of Myelinopathy in Sprague-Dawley Rats

Abstract: It is well established that hexachlorophene, which is used as an antibacterial agent, causes intramyelinic edema in humans and animal models. The hexachlorophene myelinopathy model, in which male Sprague-Dawley rats received 25 to 30 mg/kg hexachlorophene by gavage for up to 5 days, provided an opportunity to compare traditional neuropathology evaluations with magnetic resonance microscopy (MRM) findings. In addition, stereology assessments of 3 neuroanatomical sites were compared to quantitative measurements … Show more

“…Stereology is the method of choice for quantifying lung structure in microscopy, for which standards, principles, and practical applications have been rigorously defined ( 20 25 ). The same principles apply to CT, micro-CT, PET, MRI, and other modalities (“ in vivo morphometry”) ( 26 ), used in the brain ( 27 30 ), heart ( 31 ), and lung ( 26 , 32 , 33 ), among other organs.…”

“…X-rays and MRIs) (Roberts et al, 2000) to more objectively diagnose disease and evaluate imaging diagnoses and response to treatment. Potential use of qualitative and quantitative assessments of potential white matter neurotoxicants in nonclinical toxicity studies (Sills et al, 2020). Using volumetric analysis of magnetic resonance microscopy (MRM) and stereology adds significant value to standard 2D microscopic evaluations.…”

“…MR Histology is becoming much more routine in a wide range of studies in the rat brain ( Calabrese et al, 2013 , 2014 ; Johnson et al, 2014 ; Sills et al, 2004 ; Calabrese and Johnson, 2013 ; Antonsen et al, 2013 ; Sills et al, 2020 ). The new template and associated labels provided here will facilitate expanded applications.…”
