Quantification of Lipoprotein Uptake in Vivo Using Magnetic Particle Imaging and Spectroscopy

Abstract: Lipids are a major source of energy for most tissues, and lipid uptake and storage is therefore crucial for energy homeostasis. So far, quantification of lipid uptake in vivo has primarily relied on radioactive isotope labeling, exposing human subjects or experimental animals to ionizing radiation. Here, we describe the quantification of in vivo uptake of chylomicrons, the primary carriers of dietary lipids, in metabolically active tissues using magnetic particle imaging (MPI) and magnetic particle spectroscop… Show more

“…Similar to when SPION contrast agents were first realised for MRI in the 1980s, 213 research on the development and synthesis of monodispersed novel MPI-tailored SPIONs has recently become an important area of research. 5,63,189,[214][215][216][217] These new particles can be functionalised and optimised for improved performance in specific applications, like for increased circulation time or for more efficient cell targeting. Whilst MPIoptimised tracers will have to undergo a lengthy evaluation before clinical approval, the exploration and development of better performing SPIONs should spur further work towards clinical applications.…”

“…These impressive properties of Zn-ferrites in MPI have been echoed in other studies in the literature. 191,214,216 Other ferrite nanoparticles that have been investigated for their MPI properties include Ni-and Mn-ferrites. Irfan et al synthesised small Ni-ferrite nanoparticles (9-12 nm), functionalised with either a citric acid (CA) or polyacrylic acid (PAA) coating.…”

“…Being the radioactive isotope labeling prejudicial to the human or experimental animals, Hildebrand et al 194 reported a new effective strategy to in vivo quantify lipoprotein uptake focused on brown adipose tissue (BAT). It was demonstrated that the chylomicrons (primary carriers of dietary lipids), when artificially loaded with Fe 3 O 4 , enabled rapid and highly sensitive in situ using magnetic particle spectroscopy (MPS).…”
