QUANTITATIVE DETERMINATION OF HELLEBRIN IN THE RHIZOMES AND ROOTS OFHELLEBORUS PURPURASCENSW. ET K.

Abstract: Using two photocolorimetrical methods described in the literature the authors carried .out a sequential study for the quantitative estimation of hellebrin in Helleborus purpurascens. The defating of the.plant material in a Soxhlet was faster and more reliable. The drug may be considered exhausted by three extractrions with methanol as solvent and a drug-solvent ratio of 1 :20. The method indicated by the literature and based on a chloroform-methanol extraction of the first hydromethanolic extractive solution w… Show more

“…Hellebrin was isolated (at the Dipartimento di Scienze Chimiche, Università di Napoli Federico II , Italy) from Helleborus purpurascens according to a modified procedure from Cioaca and Cucu [20]. Gitoxin (ASB-00007232-005) was obtained from ChromaDex Inc. (Miami, FL).…”

“…After the homogenization was repeated three times, the petroleum ether was evaporated to obtain plant material free from oil (Wissner and Kating, 1971). The powdered plant material was extracted with 20 fold the substance's weight (w/v) methanol in soxhlet apparatus to extract the components soluble in alcohol (Cioaca and Cucu, 1974). Aqueous extraction of powdered plant material was done with its 8 fold weight boiling distilled water (Zhang et al, 1999).…”

“…The traditional and official medical use of Helleborus species is based mainly on the chemistry of some components as genetic resources. Among them, hellebores are rich in structurally diverse active compounds that are responsible for a variety of pharmacological effects (Cioca and Cucu 1974;Milbradt et al 2003;Szabó 2005), e.g. cardiac glycosides, steroidal saponins, ecdysones, and protoanemonin (Szabó 2005).…”

“…Since the majority of researches has focused on the chemical and pharmacological traits of hellebores (Cioca and Cucu 1974;Stochmal et al 2010;Yang et al 2010;Vitalini et al 2011;Milbradt et al 2003), and only a limited number of papers addressed structural issues (Vesprini et al 1999(Vesprini et al , 2012Š ušek 2008;Barkyna and Churikova 2014;Rottensteiner 2016;Kumar and Lalitha 2017), our work aimed at highlighting variability in the anatomy of three Helleborus species. Although the tissue structure of the vegetative organs and the flower-comprising peculiar coloured petal-like sepals, petals modified into nectaries, and inner rings of stamens and pistils-is fairly constant within the genus (Tamura 1993;Dános 2006), there might be slight differences regarding the morphology and anatomy of stem, foliage leaf and floral organs in various species.…”
