Quantitative Determination of the Binding Capacity of the Sex Hormone-Binding Globulin, Using Agar Gel Electrophoresis

Abstract: An easy, rapid and precise technique for the determination of the binding capacity of the sex hormonebinding globulin (SHBG) is described. The plasma is incubated with [ 3 H]5a-dihydrotestosterone (DHT) either alone, or together with an excess of unlabelled DHT. The SHBG-binding peaks are demonstrated by agar gel electrophoresis according to Wagner ) Hoppe-Seylers Z. Physiol. Chem. 353,1235-1245, and the SHBG-binding capacity is calculated from the radioactivity specifically bound to the SHBG peak, corrected f… Show more

“…FAI normally increases as a function of the exceeded SHBG saturation limit (Krieg & Arning, ; Dunn et al ., ) and was markedly elevated compared to S‐T and Free‐T in GD FtM. This might indicate a higher bioavailable testosterone fraction maybe more accentuated in GD FtM (Table ), even though earlier studies have shown that FAI is not a reliable parameter of Free‐T (Kapoor et al ., ; Vermeulen et al ., ).…”
