Quercetin protected isolated human erythrocytes against mancozeb-induced oxidative stress

Abstract: Mancozeb is a fungicide belonging to the ethylene-bisdithiocarbamate group and is widely used in agriculture. The aim of this study was to examine the protective effect of quercetin (QRN) against oxidative stress induced by mancozeb in human erythrocytes. In order to verify this, 5 ml of venous blood was collected and the erythrocytes were separated and divided into equal parts. One part was incubated with different concentrations of mancozeb (0, 10, 30, 100 µM) for 4 h at 37°C. The other part was preincubated… Show more

“…Some reports confirmed this ROS, in human mesencephalic cells and mouse granulosa cells, after mancozed treatment, since the level of oxidative stress related gene HSPA1A, HSPA1B, NOX5, SOD2 and CAT were up-regulated, leading to oxidative stress response [7]. In addition, anti-oxidant supplement would protect from mancozeb induced toxicity [39, 40], suggesting and confirming that mancozed toxicity is not only due to its estrogen-like effects but also resulting from the cell damage related to oxidative stress. Resveratrol improved mouse oocyte antioxidant system, which has been suggested previously [41] .…”

“…HSP genes are known as “stress genes,” serving as indicators of the cellular toxicity of different environmental stressors [ 47 50 ]. In this study, larger relative steady-state levels of the specific mRNA of Nrf2, catalase, HSP70, and 83 were observed at the concentration of 5 mg/mL, suggesting an adaptative response of flies to the presence of the lower concentration of MZ in the diet for 15 days [ 42 , 43 ]. In contrast, at a higher concentration, a decrease in the relative steady-state level of the mRNA of those genes occurred, and this inhibition was not extended to the activity of catalase, which remained augmented.…”

“…MZ stimulated GST activity; this effect was also described in rat hepatic tissue and carp brain [ 16 , 41 ]. The family of protein GST plays multiple roles including cell protection against oxidative stress and potentially toxic compounds via conjugation of glutathione (GSH) to various molecules and products of oxidative metabolism [ 42 ], decreasing the toxicity and contributing to their elimination from the cell. An augmented GST activity was observed in both concentrations of MZ and occurred in parallel with decreased and reduced glutathione, indicating the activation of the detoxification mechanism mediated by GST in flies.…”

“…Carboplatin in the form of PCL nanoparticles and free drug were evaluated for their potential to induce haemolysis in rat red blood cells (RBCs) as reported before [15]. In brief, rat RBCs were separated from fresh rat blood by centrifugation at 1500 rpm for 15 min to remove plasma and other cell debris.…”
