Radical scavenging activities of peptide from Asian clam ( Corbicula fluminea ) and its protective effects on oxidative damage induced by hydrogen peroxide in HepG2 cells

Abstract: The soft part of Asian clam (Corbicula fluminea) was hydrolyzed using trypsin and the hydrolysates were separated by ultrafiltration using the membrane with molecular weight cutoff of 10 and 5 kDa. Three fractions (F10000, F10000–5000, and F5000) were obtained. The F5000 showed the strongest scavenging abilities to DPPH (2,2‐diphenyl‐1‐picrylhydrazyl), superoxide anion, and hydroxyl radicals, and was further fractionated into four fractions (component I, II, III, and IV) by gel chromatography. The protective e… Show more

“…Reactive oxygen species such as H 2 O 2 may change the structure of biological macromolecules, leading to cytotoxicity and damage. Oxidative stress and the increase in reactive oxygen species are key factors of aging [ 34 ]. As H 2 O 2 is the main precursor of highly reactive oxygen species, this study used it to induce oxidative damage of HepG2 cells.…”

“…For example, Xu et al reported that hydrolysate was prepared from Asian clams (Corbicula fluminea) hydrolyzed by trypsin, and the peptide component separated from the low molecular weight part (<5 kDa) showed a significant protective effect on HepG2 cells with H 2 O 2 -induced oxidative damage. Such a positive effect was mainly attributed to the radical scavenging capability [50]. In addition, Hu et al reported that the antioxidant peptides from grass carp (Ctenopharyngodon idellus) scale gelatin had a protective effect against the HepG2 cells' oxidative damage induced by H 2 O 2 , which significantly promoted HepG2 cells growth and inhibited cell apoptosis [51].…”

“…In particular, the medium was changed every 2-3 days. (25,50,100,200, 400 or 800 µg/mL) for 4 h. In particular, after performing filtering operations with 0.22 µm spin filters, H 2 O 2 and MPs (or CPs) dissolved in DMEM medium were simultaneously added to the liquid mediums. The cell viabilities of all the above-mentioned samples were calculated, which was used to evaluate the effects of MPs and CPs on H 2 O 2 -induced oxidative damage in HepG2 cells.…”

“…antihypertensive [15], anti-inflammatory [10,11,16,17], hepatoprotective [8][9][10][11], hypocholesterolemic, and hypolipidemic effects [9,[17][18][19][20][21]. e composition of freshwater clam extract, which includes free amino acids, peptides, proteins, carotenoids, phytosterols, and polysaccharides, has been reported in previous studies [13,14,[20][21][22][23][24][25]. Abergel et al reported that palmitic acidinduced HSC deactivation resulted in suppressed cell growth and reduced the expression of both a-SMA and collagen I in the rat hepatic stellate cell line PAV-1 [26].…”

“…Freshwater clam extract alleviates nonalcoholic fatty liver disease, ameliorates exhaustive exercise induced liver, and improves hemorrhageinduced acute liver injury [9][10][11]. In addition, a number of studies have found that freshwater clam extract possesses various biologically active properties, including anticancer [12][13][14],…”
