Radioimmunoassay for Plasma Antidiuretic Hormone

Abstract: Antibodies to [8-lysine]-vasopressin (LVP) were produced in rabbits by a macrophage harvesting technique. Using these antibodies a radioimmunoassay for vasopressin was established using LVP as standard and for labelling with 125I. The antibody demonstrated complete cross-reactivity to [8-arginine]-and [8-ornithine]-vasopressin but concentrations of oxytocin 1000 times greater caused no displacement. A simple extraction procedure for antidiuretic hormone (ADH) in plasma is described. Employing this extraction p… Show more

“…DISCUSSION The development of a clinically useful radioimmunoassay for plasma vasopressin has proven to be more difficult than for many of the other polypeptide hormones. Although a number of other laboratories have succeeded by a variety of methods in raising antisera to vasopressin (7)(8)(9)(10)(11)(12)(13)(14)(15), none has been sufficiently sensitive to detect the hormone at the low concentrations (10-M) normally present in plasma. Moreover, when carefully studied, several of these antisera have been found to be unusually susceptible to interference by a The effect of acute hypotension on plasma AVP in a healthy subject.…”

“…Initially a dose of arginine vasopressin (AVP) (Ferring1 mu 2-3 pmol) was calculated on the basis of work of Johnston [1972]. Assuming that the clearance rate per kilogram body weight was the same for adult and fetal sheep, a dose of 200 pmol .…”

“…Blood pressure was measured in conscious rats by a tail plethysmographic method. Plasma renin was measured by a competitive antibody method employing radioimmunoassay for generated angiotensin I (Johnston et al, 1969 and1972) and plasma angiotensin II on extracted plasma by radioimmunoassay. Plasma vasopressin was measured at 28 days or longer after renal artery steuosis by radioimmunoassay (Johnston, 1972 ;Pnllan et al, 1977).…”
