Raised Serum IgG4 Levels in Patients with Atopy and Filariasis: Application of an Automated Particle-Counting Immunoassay Using Monoclonal Antibody

Abstract: We show here an automated (50 samples/h) assay for serum IgG4 having a throughput time of 40 min per sample and a sensitivity of 10 μg/ml. The assay procedure is based on the inhibition by sample of the agglutination reaction between monoclonal anti-IgG4 antibodies and latex particles to which IgG4 myeloma protein has been coupled. Assay reliability was ascertained by testing for linearity, analytical recovery (96.4%), interassay precision (≤8%), specificity and correlation between the results obtained with mo… Show more

“…A significant positive correlation between IgE and IgG4 has previously been found among non-atopies [15], patients with helminthiasis [15.23] and recently also among atopic adults [C. G. M. Magnusson, in preparation]. This parallelism in IgG4 and IgE responses may be governed genetically in many ways, but it is tempting to ascribe this to the influence of IL-4, a T-cell-derived interleukin, which has recently been shown to afl"ect IgE and IgG4 synthesis by human lymphocytes in vitro [24].…”

“…An increase in the production IgG4 during infection with helminths has been previously described (Ottesen et al, 1985;Magnusson et al, 1986). However, increased IgG4 antibody levels has been generally associated with chronic antigenic stimulation (Magnusson et al, 1986;Aalberse et al, 1983).…”

“…In fact, relationships between serum concentrations of IgE and IgG4 were observed preferential ly in individuals with very high levels of IgE; in patients with helminthiasis [ 12] and in infants whose mothers were atopic [13]. So, allergic patients (asthma/rhinitis) presented concentrations of IgG4 not significantly higher than in con trols and not correlated to IgE concentrations.…”
