“…After DCRBF was dissolved in ethyl acetate, the polymer (PLA) was removed by ethanol precipitation. The extracts were dissolved in ethyl alcohol, and the concentration of DOXY was measured with high-performance liquid chromatography (HPLC) [39]. Measurement data were expressed as a percentage of the DOXY volume to estimate the DOXY originally contained in the DCRBF.…”

“…To verify the side effects of DCRBF on the implanted mice, the DOXY blood concentration was measured by HPLC [39]. In addition, a complete blood count, liver function examination (serum concentration of glutamic oxaloacetic transaminase; GOT, glutamic pyruvate transaminase; GPT, and gamma-guanosine triphosphate; g-GTP), renal function (serum concentration of blood urea nitrogen; BUN and creatinine; Cre), and pancreatic enzyme (amylase; AMY) were also performed in these mice at each time point, and the results were compared with those of normal mice.…”

“…Total doxycycline concentrations in plasma and sputum were quantified by validated assay procedures using reversephase chromatography with UV detection as previously described (19). Briefly, plasma samples (500 l) were spiked with the internal standard (500 l of piroxicam at 5 g/ml in acetonitrile).…”

“…To increase the reproducibility of Cre-mediated recombination, we recently re-examined the inducible conditions with intravitreal delivery of doxycycline (Dox), a tetracycline derivative ( Fu et al 2014), at a concentration (4 µg in 1 µl) that does not cause retinal degeneration (Chang et al 2000). Intravitreal injection raised the retinal Dox concentration to ~ 250-fold that of the maximal level in the bloodstream delivered by feeding or intraperitoneal (IP) injection (assuming that the diameter of a mouse eye is 3 mm) (Ruz et al 2004). As feeding or IP injection depends on the blood circulation to deliver Dox to the eye, the relative retinal Dox concentration delivered by intravitreal Dox injection is likely much higher than that from feeding or IP injection.…”
