Rapid detection of pork DNA in food samples using reusable electrochemical sensor

Abstract: This study aims to demonstrate the potential use of reusable electrochemical sensor for detecting pork DNA in solution. The approach was based on electrochemical principle in which the electrostatic interaction between DNA and redox species will generate detectable signal upon introduction of electrical charge. In this study, Ruthenium Hexaamine (RuHex) was used as the redox species and result was based on the output current. Coupled with highly specific Polymerase Chain Reaction (PCR) primers designed for por… Show more

“…Some of the isothermal amplification methods that have been introduced are loop-mediated isothermal amplification (LAMP) [34][35][36] , helicase-dependent amplification (HAD) 37 , isothermal strand displacement amplification (SDA) 38 , rolling circle amplification (RCA) 39 and signal-mediated amplification of RNA technology (SMART) 40 . Among all these techniques LAMP appeared to be popular for pathogen detection as it is capable of amplifying medium-to long-range of targeted nucleic acid strands with high efficiency, sensitivity, specificity and stability [41][42][43][44] . The combination of LAMP in biosensors allowed for a straightforward detection of pathogen using paper-based colorimetry assay, and this approach could be useful as POC devices 45 .…”

“…To address such limitations, researchers have developed isothermal amplification processes like loop-mediated isothermal amplification (LAMP), 86 , 87 , 88 rolling circle amplification (RCA), 89 nucleic acid sequence-based amplification (NASBA) ( Table 13.1 ). 90 These isothermal amplifications, especially LAMP reaction, reduces time, multiple steps, costs, and development on-site based on rapid detection ( Fig.…”

“…Furthermore, the antigens of interest must be stable at high temperatures to endure the harsh conditions of food processing [4]. Over the years, numerous researchers have presented different techniques, including electrochemical-based and paperbased techniques using DNA molecules to identify animal species in foods [5][6][7]. Among the developed DNA-based techniques, polymerase chain reaction (PCR) methods have been used extensively for food authentication purposes.…”
