Rapid determination of nucleotides in infant formula by means of nano‐liquid chromatography

Abstract: A rapid method for the quantification of five ribonucleotides 5'- monophophates (adenosine, cytidine, guanosine, inosine, uridine, 5'-monophosphate), in infant formula, has been proposed using nano-LC. To separate the studied compounds, capillary columns packed with different C18-based stationary phases were investigated. All the columns tested were laboratory prepared. The experiments were performed in ion-pairing RP chromatographic mode using tetrabutylammonium hydroxide as ion-pairing reagent. The method wa… Show more

“…The analysis started immediately after positioning the injector device in the injection mode. When the mobile phase had to be changed, it was directly introduced into the modified injector, significantly reducing the consumption of organic solvents [34]. The flow rate was estimated by connecting a microsyringe of 10 µL (Hamilton, Reno, NV, USA) to the capillary column end through a Teflon tube and by measuring the volume of mobile phase collected for 5 min.…”

“…The nucleosides are trapped and concentrated on the pre-column, and then they enter the analysis column for separation, which is caused by gradually changing the elution solution [ 103 , 109 , 110 , 111 ]. The column stationary phases are also critical to separation [ 112 ], and are often replaced with novel materials that interact with nucleosides for online enrichment and improvement of separation [ 73 , 113 ].…”

“…The analysis started immediately after positioning the injector device in the injection mode. When the mobile phase had to be changed, it was directly introduced into the modified injector, significantly reducing the consumption of organic solvents [73]. In the optimized conditions, the flow rate of the column was about 450 nL/min.…”
