Rational Design of Multifunctional Gold Nanoparticles via Host–Guest Interaction for Cancer-Targeted Therapy

Abstract: A versatile gold nanoparticle-based multifunctional nanocomposite AuNP@CD-AD-DOX/RGD was constructed flexibly via host-guest interaction for targeted cancer chemotherapy. The pH-sensitive anticancer prodrug AD-Hyd-DOX and the cancer-targeted peptide AD-PEG8-GRGDS were modified on the surface of AuNP@CD simultaneously, which endowed the resultant nanocomposite with the capability to selectively eliminate cancer cells. In vitro studies indicated that the AuNP@CD-AD-DOX/RGD nanocomposite was preferentially uptake… Show more

“…Therefore, various nanoparticles based drug delivery systems have been studied [ 2 , 27 , 29 ]. The targeting of RGD-modified nanoparticles to tumor vasculature is a promising strategy for tumor-targeting treatment, because some tumor cells within solid tumors would overexpress α ν β 3 integrin, which can specifically recognize the peptide RGD [ 20 , 23 , 36 , 37 , 38 ]. Buckley and co-workers [ 39 ] described that RGD-containing peptides enter cells and directly induce autoprocessing and enzymatic activity of procaspase-3, a pro-apoptotic protein.…”

“…In addition, Dox-encapsulated nanoparticles formed by poly(RGD- co -β-amino ester) and Dox-polymeric nanoparticles functionalized with RGD had higher cytotoxicity than nanoparticles prepared without RGD in U87MG cells [ 24 ]. Besides these, in recent years, other authors have described the improvement in transport of Dox into tumor cells with targeting of RGD-nanoparticles using different materials and methods of preparation [ 37 , 49 , 50 , 51 , 52 , 53 ].…”

“…Currently, available chemotherapeutic agents are nonselective and usually have side effects. It is expected that functional nano-based drugs such as magnetic, gold, and silver NPs will overcome this nonselectivity and will improve and enhance the result of cancer therapy [87,88].…”

“…Gold nanoparticles (AuNPs) have been widely used in drug/gene delivery [1,2], cancer-targeted therapy [3], photothermal therapy (PTT) [3,4] and diagnostic imaging [5], due to their high stability, relatively easy synthesis, chemical inertness and facile ability to combine ligand. Gold is generally considered to be chemically inert and biocompatible, but due to its small size and large surface area, AuNPs may have higher bioactivity than bulk gold.…”
