Reactive Oxygen Species Formation in the Brain at Different Oxygen Levels: The Role of Hypoxia Inducible Factors

Abstract: Hypoxia inducible factor (HIF) is the master oxygen sensor within cells and is central to the regulation of cell responses to varying oxygen levels. HIF activation during hypoxia ensures optimum ATP production and cell integrity, and is associated both directly and indirectly with reactive oxygen species (ROS) formation. HIF activation can either reduce ROS formation by suppressing the function of mitochondrial tricarboxylic acid cycle (TCA cycle), or increase ROS formation via NADPH oxidase (NOX), a target ge… Show more

“…According to several studies, cell injury, as well as cell death is mediated by hypoxia through apoptosis, oxidative stress, acidosis, and inflammation (33) . On the other hand, several other studies showed that apoptosis is induced by hypoxia through the inducement of calcium overload, increased oxygen free radicals, mitochondrial damage, and the increase in HIF (hypoxia-inducible factor) expression (34,35) . As a result, hypoxia is closely related to apoptosis and this is concur with our investigation.…”

“…Historically, a variety of acute and chronic O 2 ‐sensing mechanisms have been proposed. These include increases, or paradoxically decreases, in reactive oxygen species (ROS), activation or inactivation of metabolites, for example, nitric oxide, prostanoids, etc, AMP‐activated protein kinase, ion channels, Na,K ATPase, haeme and other proteins and the hypoxia inducible factors …”

“…Following the metabolic modifications and linked to the oxidative stress, ROS (Reactive Oxygen Species) can increase dramatically and lead to important DNA damages, aiming to destroy tumor cells [62]. The ROS in brain tumors has long been studied since they are major criterion for radiotherapy resistance.…”
